You will need to wipe both drives, Wipe -> Create the Raid -> Format (create file system)
A better alternative is to use the second drive for rsync or rsnapshot from the first, in essence a mirrored copy.

What's secure as far computer hardware is concerned neither are secure, do you understand mdadm (OMV's software raid) if you get a problem could you resolve it from the command line.
1 drive for data, second drive for rsync or rsnapshot, which in effect backup the first drive, either are run as scheduled jobs usually in the early hours and with email notifications to tell you if the job completed or not
You could also create a RAID-1 Array with one drive missing, copy the data from the 1st disk to the array, wipe the 1st disk, add it to the array and let it sync.
Thank you all for your replies. I think I'll create a rsync job to copy my data 1 or 2 times per day, it seems the "simplest" way if one of my HD dies.
I i'm also considering to use my two 2 tb hdds as 1 data hdd and 1 as backup with ext4.... but if i run out of space, can i later buy 2 other disks and extend them in any way (like 2 6 tb master pool and 2 6 tb backup) and continue using rsync? or would be better a raid-0 right now on master disc? excuse me, i'm also a newbie...
You can use mergerfs. In OMV this is the unionfilesystem plugin. With this you can have multiple drives under the same mount point. So for example two drives will look like one.
This is used under the hood and the OMV plugin provides the GUI
